Quantifying HeLa cells in a 96 well plate - (Jan/19/2007 )

Hi All,

I just recently started working with HeLa cells and one of the experiments I am trying to conduct requires non-invasive quantification of HeLa cells in several of the wells in a 96-well plate. I can not take them out or use dyes. Can anyone suggest a good strategy?

As well, what would be the approximate number of cells in a well, when the substrate is fully covered (i.e. what is the approximate cell density per cm^2 when the polystyol substrate is completely covered with HeLa)?

Take a pic of the cells (as they r in the 96 well plate) and count them manually or may b some program could count for you.

Number of cells/mm2 depends on cell types. i havent worked with Hela cells, so no idea.

Thanks for your help. However, since the cells are not stained and the area of each well is 0.3cm^2, it will be fairly difficult to photography and quantify.
Can you suggest any software for counting non-stained cells in a picture?
