How to import GeBank files into Primer Express 2.0 - (Jan/18/2007 )

I would likr to develop qPCR primers using the progran Primer Express 2.0 from Applied Biosystems. These primers should cover the intron-exon border.

According to the manual, primer Express can find the intron-exon borders from an imported sequence. But only if a GenBank file is saved as an unformatted text file.

However, if I want to save a Genbank page as a text file, a also have to choose an encoding option (zie attachement). Which option I choose, Primer Express will not recognize the file.

Copy/paste to WORD also didn't work.

So what"s the best way to create an unformatted text file that is recognized by Primer Express 2.0??

(I'm working with an official CD primer express, not an illegal copy)

Theo22, I havent' use primer express, but I think your question is much of a general question of creating a genbank plain text file.

Go to ncbi, display you sequence under genBank, then from the "send to" drop down menu select text, now you will get a unformatice flat text genbank file for your sequence, then copy it and paste in a text editor on you PC/mac, then save it. Now, if you are using a pc, you should save it as .txt file, while if you are using a mac, be sure to specify you are saving it as a flat text file without any formatting.

Just copy stuff of your browser won't work, because when you paste it in word, it automatically remember all the links and html formats. that why you got to "send to" text format then copy it off your browser.
