Medium preparation L-15 for MDA-MB-231 - (Jan/08/2007 )

Hi,

Have a quick question for ya'll. We've been trying to culture MDA-MB-231 breast cancer cells in L-15 medium but for whatever reason they seem to be growing exceedingly slow. We had them at one point and were subculturing at a ratio of 1:10 twice a week so this is definately not normal. We've since had to make up a new batch of L-15 medium and I was curious if we had somehow left a constituent out. We've been using mediatech L-15 medium (50-045-PB) with L-glutamine added, reconstituted in water, with 10% FBS. No other supplements were added.

We grow our cells in a 5% CO2 incubator. But with this medium we have the flasks capped tightly.

Leibovitz's L-15 medium is formulated for growth of cells in CO2 free system. You are trying to culture these cells in CO2 incubator, just grow them in humidified incubator without CO2.

i grow my mb-231 in RPMI with 10%fbs, hepes, and pen strep glut. never seen a problem with them in that media.

I've always grown them in RPMI 1640, 10% FBS, 2% P/S, 1.25% L-glut, and 1% sodium pyruvate in a 5% CO2 incubator without any problems.
