Selection of cell culture media - for HT29 colon cancer cells (Jan/04/2007 )

I have searched PubMed for the good culture media for HT29 cells. More articles I reviewed, more I confused. A range of media including DMEM, MsCoy's 5A, RPMI 1640, MEM were extensively used. Moreover, some of them added glutamine and other nonessestial aminoacids and others didnot. One's media contains 3.5g/l glucose and others has 4.5. L-glutamine concentration also varies from one to another.

I wanna do western blotting for some Apoptosis signalling proteins and MAPK using HT 29 cells, could you please help to select the right cell culture media.

Some cell lines are really fussy about the media you grow them in but HT29 don't really care, so don't worry too much about it. If you're getting these cells right from a supplier like ATCC you're probably best to go with the media they recommend, as they will be adapted for growth in that media; if not, just make a sensible choice.
I usually grow them in Gibco's DMEM (+GlutamaxII) supplemented with 10% FBS and 4 mM L-glutamine (even if the media is supposed to contain L-glut it's so labile I usually add it in anyway). Works just fine.
