isolation of streptococcus faecalis - (Dec/15/2006 )

hello
i am trying to isolate s.faecalis from yoghurt.none of the media that i have tried till now have worked......the main problem being overgrowth by lactobacillus.have tried aerobic conditions, high ph(9.6)...but have not been successful.
can anyone suggest any medium as well as growth conditions?

Strep. faecalis is now know as Enterococcus faecalis.

As fas as I remember there selective media for E. faecalis when it's isolated form the gut.

The media descripbed in the following pages might point you in the right direction
http://service.merck.de/microbiology/tedis...05262_0500.html
http://www.pubmedcentral.nih.gov/articlere...ubmedid=4214072

It would also be good to tell us which type of media you used.

thanks for the links.it will be of much help. rolleyes.gif
the medium i used is my own modification of chesbro and evans (1959).......proteose peptone 1%,yeast extract0.5%,glucose 10%(autoclaved separately),dipotassium hydrogen phosphate 0.1%,NaCl 0.25%,azide 0.025%,agar 1.5% pH 9.6.
the azide concentration and the alkaline pH wasnt successful in preventing lactobacillus overgrowth.today i have inoculated from serially diluted yoghurt into lactic bacteria differential agar(mc donald et al AEM 1987,53:1382)) with some modifications .heterofermentative enterococcus faecalis should grow as bluish green colonies in the next 24 hrs......am i on the right track or is this gonna be my roadblock
