Dissolving Type I collagen - (Dec/12/2006 )

Hi. I have bovine achilles tendon collagen I which i have added to 50mMTris-HCl (ph7.5), with 5mM CaCl2 , to a final concentration of 1mg/ml. It doesn't dissolve. Does any one know how to dissolve type I collagen?

i have wokred with calf tendon type I collagen before,

you will have to dissolve it in 0.2M acetic acid, on a stirrer at 4 degrees, time taken to dissolve will depend on amount of collagen you have.

then prior to use you need to neutralise it with sodium hydroxide to an acceptible pH

I followed the same kind of protocol for collagen I extraction from rat tails tendons. After stirring at 4°C, I centrifuged my solution at 14000rpm 4°C for 1 hour to discard waste.
