transfecting adherent cells - before adherence? - (Dec/08/2006 )

Has anyone ever tried transfecting adherent cells, before they adhere? That is, I would like to transfect some HEK cells, but my plates are a little too confluent (about 80%: I prefer 50%). Is there a way I could trypsinize them, then add the transfection reagents, then plate them? unsure.gif

Yer, there is the retrotransfection method with lipofectamine or lipofectamine2000. It is on the datasheet. An alternative is to seed them in the morning, wait for 5 hours ca. and then transfect them.
