Stable cell trasfection questions... - Integration of plasmid... (Dec/06/2006 )

Hi, there.

Some basic questions on stable cell transfection... In which moment the plasmid is incorporated to cell DNA? What enzimes participate? Is it a kind of recombination process? The site of incorporation... is it random?

I have the same problem for selecting a stable clone from primary neural stem cell.

I tried to select the clone from neural stem cell that expressing my gene but it lost the

differentiating ability soon after the selection. I have tried many times as well as got the

same results. It was really tried to pick the spheres from the bulk cultures under

microscope !!

Transfection reagent : DMRIEC & Lipofectamine 2000
