ROS and NO detection using fluoresceins - (Nov/29/2006 )

Pls,
would somebody provide me with protocol how to measure ROS and NO (separately) in 96 well plate? I have no signal even with controls 0,5 mM DMSO a 1M Na nitroprusside. I have filters ex485/510 em538 but according to publications people are using them even if they are not optimal. Is it possible to load cells with the dyes by adding them into medium? Is possible to measure fluorescence in DMEM with fenol red? Any reply is welcome. Thank you.

Do you use DAR-4M AM?
For NO I would say, threath the cells with SNAP, you're sure that your NO is high and than try. And I would first use fluorescent microscopy to detect fluorescence before going to plate readers.
