Can any one help me with restriction digestion map for DNA? - (Nov/18/2006 )

A DNA has been cut with Enzyme 1 and Enzyme 2.

DNA has been cut by Enzyme 1 at 8kb, 6kb and 3kb.

DNA has been cut by Enzyme 2 at 10kb and 7kb

DNA has been cut by both Enzyme 1 and Enzyme 2 at 8kb, 6kb, 2kb and 1 kb


Can some one explain to me how to create a restriction digestion map for this DNA?

Oh and pleeeeease explain it step by step! I am finding it difficult to understand this!

First of all, I did a draft showing the DNA as linear because it was easier to draw it.
The one you see in figure, is the only way of putting the pieces together...

Now i understand the map for linear dna. now i need to undersrand the map for circular DNA sad.gif

It is the same, if you consider that the first and the last cut of each linear DNA are only one cut. Just think you are cutting sequencially with each enzyme (the enzyme cuts the first time, you have linear DNA and then you go on)
