Hi Dinesh,

I think the cell no. that you are using is high for such a fast growing cell line. Could you reduce it to 10 or 15 thousand cells per well.

Final DMSO concentration at 0.5 to 1.0% is okay.

Many thanks for your kind suggestions.
Regarding DMSO, I have been using final DMSO concentration 2% (2+2 ul/200 ul medium), coz we are testing significance of co-treatment. I dont know this has been a problem to me???

Hi Dinesh!
I agree with exploresci, there are too many cells in the wells. How long does it last your treatment? If you want to see several times of exposure (i e 24h, 48h up to 144h) maybe you should start with 5000-8000 cells per well.
About DMSO why don't you leave a row/column of the plate to test just DMSO 2%? so you can compare it with your control.

I have modified my ongoing (repeat) experiment according to guidelines of you all specially exploresci.

For you information, exposure is 24 hrs(regarding drug treatment)

Furthermore, exactly what you told, I did the same (to test the 2% DMSO).

This time I am hoping best (coz you all guys with me).
