3T3-L1 cell differentiation - (Nov/10/2006 )

Hey guys,

I am trying to differentiate the 3T3-L1 cell using insulin. I have the paper describing how to do it and after differentiate you need to do assays to confirm differentiation. The Oil Red O staining is carried out on cells that grow on the eight-chamber microscope slides.

What is this slides? where can I buy? and when do u transfre the cell to this slides? I understand that I start with a 60mm dish cell, just change the medium containing insulin...everything needed to induce the differentiation. And after around 10days, perform assay. But if this staining assay is carried out on the slides, I need to trypsinize the cell and grow them on the sildes before I stain them?

Could anybody give some hint about this?

I'm not sure whether you got your answer during this period. first of all, the chamber slide is a glass slide with a plastic chamber on top which you can use it for culture your cells, and by removing the chamber then you can cover it with a cover slips. It is a gadget for those who wish to do immunostaining(or might be other usage). so if you want to use the chamber slide for your adipocyte induction, you'll have to seed your cells on the slide form the beginning. you can check NUNC website for the chamber slide. good luck !

I believe that you can replace this expensive device with simple coverslips. Just place one autoclaved coverslip on the bottom of your 60 mm dish and grow cells as usual, then take the slip and stain them.
