G418 conc for HEK293 cells - (Nov/08/2006 )

In my experience, I use 1000ug/ml for hek293 cells. Plate the cells in g418 containing medium. You can see them stop growing the next day. Alternatively, using puromycin is much easier for selection of hek293 cells.

I'm using 0.4mg/ml and it works fine. For about a day it will look like nothing is happening but then your control cells (assuming you have one) will start to look miserable and start dying a few days after that. In your transfected cells you will notice a lot of dead cells as well but after a few days you may notice some healthy looking cells growing underneath the debris and seems to stick to them for whatever reason. Basically be prepared to wait.

Also if your dishes were 100% confluent like mine were, then for about 3 days it will look like the cells are not dying. That's just the way it is.
