New to transfection field... can u help? - (Oct/23/2006 )

hi everybody

i am new in transfection and usually i like to read more about what am i doing not just protocols.
i started my first transfection yesterday and while waiting i got some informations from google about transfection -plasmid type- but thought of you guys if u can help.
where to start my reading and if u got some links it will be much appreciated.

You should begin with the Chapter 16 of Sambrook and Russell's Molecular Cloning Book (Vol. 3). Here explains the essentials of the technique and several protocols for transfection, i. e. lipofection, calcium-phosphate, DEAE-Dextran, electroporation, biolistics and polybrene. It includes even information panels about almost any question you could have on mammalian cells transfection.

i have a question when we dilute our plasmid or lipofectamine2000 we use media opti-MEM I THINK ITS SERUM FREE?
WHY DO WE USE SERUM FREE MEDIA?
