Dynal (Magnetic) beads for ChIP? - (Oct/18/2006 )

Hi everyone,

I was wondering if any of you out there have used Dynal Magnetic beads (such as protein A Dynabeads) to do the IP for your ChIP assay. I'm still getting some background amplification with my IgG control or beads alone control so I'm looking for anything to reduce the background. I am currently using the salmon sperm DNA protein A agarose from Upstate. If you haveused the Dynabeads, please let me know either way how it works for you.

I had little success with both ptn-A-agarose and dynabeads. But i would definately say the dynabeads were better for reducing non spcific signal. Try optimising the amout you use (i started at 5ul per IP and ended up on 2ul).

Dynabeads are nice to handle; try also Dynabeads-labeled secondary Ab; important, clear your sample before ChIP with Dynabeads-protein A/G or Dynabeads-secondary Ab
