WI38 cells - how fast do they grow? (Sep/16/2006 )

I am culturing WI 38 cells. The vial I thawed is at P3 and this cell line dosen't seem to be growing fast at later passages - say after P8 or P9. The flask takes quite long to become fully confluent for the same split ratio than earlier passages. Does anyone have experience with this cell line? Can you please share your expeirence about this cell line.

I think it is possible this happens. Are you following the guidelines from ATCC? http://www.lgcpromochem-atcc.com/common/ca...tccNum=CCL-75.1

Ive been noticing the same thing you are. I thawed a vial of WI-38 cells (which were frozen at passage 6) and had great growth until the 9th passage. After that, they pretty much plummeted and its really irriating me.

im currently researching this and trying to figure out why and if theres anything i can do to help them along. if youve got anything, please let me know :\

I have no success in increasing the growth phase for these but did find a way. What I am doing is during the fast growth phase, I split 1 or 2 flasks as a backup that would go for long time (~1 week). If necessary, I add fresh media to the flask. When the cells have reached P10, then I split the backup too. In this way, the culture is going for a long time. Also the split ratio towards the later passages is 1:2!!

By the way, what medium are you using? I use RPMI modified with 10% FBS and 4mM Glutamax.

Anyway, these cells are a problem - need constant monitoring.
