FBS weaning - (Sep/15/2006 )

I have one lot of FBS that works, but only a couple of bottles. I also have one lot that doesn't work.. had anyone ever made a gradual transition from one lot to another by mixing various ratios of the two? Is it possible to get the cells to grow in the "bad" FBS by slowly adjusting them to it?

bad FBS ? you mean its older than its shelf life or on what basis? did you check if its complment inactivated?

Dear Scientistbyday,

Chuck away the "Bad" serum if it is not supporting your particular cells. The companies that sell serum will normally send you a small amount of a couple of batches to try. This should always be done to compare the serum you are using with another new batch. Once tested buy as much as you can afford to so it is at least 2 years before you have to test again.
The companies won't like me for saying this, but in my experience the SERUM used by date can easily be stretched by another 3 years. All the companies do if they have a batch that goes out of date is to retest and relabel.
