MDCK cells for barrier tests - Who has experience? (Sep/09/2006 )

Our MDCKII cells tend to build several cysts in the monolayer. As we like to start with permeability tests, I suppose that these cysts will increase permeability which means counteracting to test barrier function. Has anyone experience with permeability tests utilizing MDCKII and cysts?

Did you trypsinize MDCK and make a good effort to get real good suspension of individual cells. I found in my studies (unrelated to monolayer filter) that if your cell suspension has clumps it will form cysts when attached.
