transgenic mouse - identification by PCR (Aug/27/2006 )

Hi,

I was wondering if anyone has some tips on designing primers for identifying a transgenic mouse?
Would you design them to cross the promoter and the gene of interest, or just the gene of interst, or somewhere else?

To genotype transgenic mice I use primers in any region of the original construct (promoter, ORF, or poly A site) that yields the least number of hits when I check their sequence by BLAST.
