ChIP crosslinking - (Aug/22/2006 )

Hi,

How do you know if you are cross linking enough? Is there a way to test this before actually doing the entire ChIP protocol?

if they are adherent cell, try trypsinization- which should not work if the cell are crosslinked. I checked this on firbroblasts only.

"Enough" can depend on what you are trying to pull down. Histones require little to no crosslinking (though crosslinking is recomended to avoid redistrubution) while TFs can require extensive crosslinking. In either case you won't be able to tell until you go through with the ChIP assay.
