gDNA extration questions - help - (Aug/07/2006 )

can you discuss the following:

An agarose gel which is loaded with a concentration series and an agarose gel which is loaded with a 100bp molecular size marker ("ladder")


Also what possible reasons may DNA be degraded after the isolation process?

Also what results would one expect from a Abalone (Haliotis midae) genomic DNA extration??

(experiment - we are aiming to isolate genomic DNA from Abalone)

Nucleic acids can degrade after extraction if they are contaminated with nucleases. If you are using sterile reagents, tips, tubes etc. you should be fine as far as DNA goes. 100 bp ladder is just a marker, a plasmid or something which has been cut to produce reference bands differing by 100 bp.

100bp marker is for you to compare your Sample so you can determine the size of the fragment. It's like a ruler. There are alot of sizes, 50bp, 1000bp etc. depends on your work.

DNA could be degraded due to nuclease like has been said by Neil and if you left it in room temperature for too long. Depends. It's best to keep at -20 if you are not using it.

Results from a Abalone, you need to search and read journals, maybe population journals on Abalone to see expected results.
