depolymerizing microtubules in 293 cells - (Jul/29/2006 )

hi, i am trying to depolymerize microtubules in 293 cells using colchicine at 100 uM for 2, 3, 12 and 24 hours but there always seems to be networks of surviving microtubules. in contrast, 10 uM colchicine for 2 hours could sufficiently depolymerize most microtubules in A549 cells. even adding a cold treatment in 293 does not seem to help much. does anyone else know of a good strategy to get rid of microtubules?

I am working on CHO-K1 cell line for the study of Chromosomal Aberration. I am using Colchicine to depolymerize microtubule to arrest the cell in Metaphase. i used to treat the culture (5ml medium) with 0.4ug for 4 hrs. I am successful to get good results and to arrest the cell in metaphase. U can try this with fresh medium. if getting any problem plz contact me
