Does it need cotransfection vector when retrovirus transfected hek293? - (Jul/19/2006 )

sad.gif ,hi,everyone,I need help,I want to transfect HEK293 cell with retrovirus vector,Does it need an extra cotransfect vector,for example,env expressing vector?on the other word,does HEK293 contains env expressing gene?

oh,I meen I want to use HEK293 as packing cell to produce retrovirus.

The original 293 does not carry env. Are you working with packaging cell lines derived from 293?

for infection by a virus, no need to cotransfect.
If you want to transfect a plasmid and ask your cell to produce retrovirus, you need a packaging cell line. 293cells are not able to carry that. But phoenix cells = 293PACK are ok

I want to choose a suitable packaging cell line to package my retravirus vector inserted by goal gene,then I let the virus to infect a leukemia cell line,someone tell me HEK293a meet my demand,but,only HEK293,ad293,pa317 is easily available,I want to know which one I can use.

U need to co-transfect with a envelope protien, gag,pol,and rev plasmids along with ur vector containing the transgene.

There r different packaging cell types, we use 293T or 293FT.

Thanks a lot,Can i use pa317 to produce retrovirus dirrectly without cotransfection?Is it suitable to infect a human cell line with virus produced by mouse cell line?

U need to co-transfect with other plasmids unless the cells stably expresses these proteins.

I guess it shouldnt matter where the virus was produced. As we used produced virus in 293 and infected mice or rat.
