Help with Jurkat sonication - (Jul/12/2006 )
Can anybody tell me how I can obtain a sonicate in jurkat cells about 300-500pb?Normally I obtaines too smear about 1kb. I sonicated 6x10" in medium power position, and I use about 15 million of cells in 400ul. Thanks

What kind of buffer do you sonicate in?

What kind of buffer do you sonicate in?
I used a buffer that contains 50mM Tris-Cl pH 8,10mM EDTA,1% SDS and protease inhibitors.
