Need protocol for RNA isolation from PBMC - (Jul/09/2006 )

Does anyone have a good protocol for RNA isolation from monocytes

I use the ficoll density gradient seperation to isolate PBMC's and then further select monocytes using CD14 microbeads. I culture 5 x 10^5 cells/well in a 24 well plate. I have not managed to extract RNA using Trizol.

I am not sure where the problem is, your help is greatly appreciated.
Also on another note does anyone know if monocytes divide (i know they differentiate).

recommend RNAeasy mini kit (if you have more than 100,000 sorted cells) or RNAeasy micro kit (less than 100000 cells) for RNA extraction from any animal cells. it is the best one i have ever used.
