Maize tissue culture - (Jun/11/2006 )

Hi,

I am new in tissue culture. I will going to do the maize tissue culture soon but I don't have the protocol for maize tissue culture. I will going to do the research on maize Agrobacterium transformation. What media I should use for callus culture initiation from maize mature seed?Anyone can give your helping hand? Thank you!

I'd like to help you, but unfortunately I don't have any experience in maize culture.

Why did you choose A. tumefaciens transformation strategy? (I have some experience in that field, so if you have some additional question...) I mean: maize is a monocot, isn't it? I thought that A. tumefaciens method is hardly appliable to monocots...am I totally wrong?????

yes ila is right try biolistic method for maiz.

That's about Agrob. transformation of maize callus...so, at the end it seems that I could have been totally wrong dry.gif ...
what do you think? Is Agrob. a suitable agent to transform monocots??? huh.gif huh.gif huh.gif

Thanks for ur help. What i know there are many successful cases transform used agro. My lab done a lots of transformation using agro for oil palm n rice (both monocot). So i dont think is a problem if use agro to transform monocot. smile.gif
