Measuring cell viability via luminescence... plate type? - Promega CellTiter-Glo kit (Jun/07/2006 )

Hello all,

I've been using Promega's CellTiter-Glo cell viability assay which uses a luciferase reaction to measure ATP levels. I'm measuring the luminescence resulting from this reaction in a 96-well opaque, black plate using PerkinElmer's Wallac Victor3V plate reader.

My question is: Does anyone have experience using non-black, opaque plates? I've always been told that you need to use black, opaque plates for luminescence measurements. Since the plates are opaque, I cannot monitor the cells during culture. I tried using clear plates and taping the bottom with black vinyl tape and it seems to be working fine. I then tried just a regular, clear plate and the results were nearly identical to those of the taped plate.
Just some interesting observations. Any feedback would be appreciated.

Wy you did not try to use a black plate with the bottom clear? In other words, only the walls of the wells are in black, while the bottom and the lid are colorless. Many company produce it and is very easy to follow the grown of the cells. I used this one with the same plates reader that you're using and the results were good, of course i used another kit. finally wy did yoo use the black plates? generally for the luminiscence strategy is better to use white plates, because the white walls don not adsorb light and the results are most sensivity. you can compare results from a white/black opaqe plate with the clear plate to further speculate, if you have the money to spare for the ATP GLO assay tongue.gif

i have used CellTiter-Glo with white opaque plates and everything seems to be perfect. to monitor the cells during or following incubation just make a duplicate in a transparent plate and follow it up or just do what amicia did!
