arabidopsis leaf tissue culture - (Apr/19/2006 )

Hi there,
Does anyone know of a way to to culture Arabidopsis leaf explants so that they form a callus / undergo somatic embryogenesis? I have successfully done this with Medicago truncatula, as described in: Nolan, Rose (1998) "Plant regeneration from cultured Medicago truncatula with particular reference to abscisic acid and light treatments". AUSTRALIAN JOURNAL OF BOTANY 46 (1): 151-160. But I have had no luck at all finding this info for Arabidopsis. I have found protocols for somatic embryogenesis from leaf protoplasts, flowers, siliques, shoots etc etc but none for leaves. If anyone knows of any protocols or has done anything like this I'd be very grateful for information. Information from other organisms may be helpful as well. Thanks!

I checked some literature databases, but I couldn't find anything specifical about indirect somatic embryogenesis in A.t. Anyway, I send to your PM box a review that seems to be intresting.....maybe searching through quotations.... rolleyes.gif
My work is in part focused on callus and embryogenesis induction from grapevine cultures, so I have a little experience in this specific field. If it could help you, I can send you some reference to experiments that use Vitis leaf explants, but I believe monocots and dicots are really too different to make a comparison...

I would like to see those Vitis leaf explant protocols if you wouldn't mind. You may be right and they are too different, but I haven't found that many leaf explant protocols so anything I can find would be helpful. Cheers!
