Caco-2 Vs Hybridoma cells - contamination of Caco-2 cell line with hybridoma cell line (Mar/29/2006 )

blink.gif hi friends ! iam handling few cell lines ! we recently bought Caco-2 cell line. i have ended up mixing caco-2 cells with hybridoma cells and now iam unable to remove this contamination. Hybridoma cell line has a doubling time of just 24hrs is growing fast and soon will replace Caco-2 cells. caco-2 cells are really slow growing cells. i tried few mwthods to isolate them but failed.
1. tried to stave to induce apoptosis but they survived for more days than Caco-2,
2. Tried to scrape with a scrapper, but they spead to other areas very rapidly.
iam really fixed,
can any one help me out to separate these cells so that i can save Caco-2 cell line.

i also tried trypsinising . as caco-2 takes longer time to detach when trypsinised i tried differential trypsinisation. but some of the colonies of hybridoma remained as it is and some started floating.
is it checkmate?????????????????????????????

In my experience, caco2 is very resistant to puromycin, while most other cell lines are sensitive.

In my experience, caco2 is very resistant to puromycin, while most other cell lines are sensitive.


Caco2 growing at high density can tolerate higher dose of puromycin. Maybe you would like to try several dosages, 1 to 10 ug/ml, for example. Usually, puromycin sensitive cells will die within 5 day.
