weird bubbles in LB plates.. - (Mar/23/2006 )
HI
After I spread my Ecoli onto LB plates, and leave them at 37C ON, I get bubbles on the bottom of my plates. Does anyone know why that might be?
thanks!
You might have poured your plates with too much warm agar.
(doesn't sound English ) In other words : the medium might have been too warm when you poured the plates.
it's not a big problem, just ennoying when you try to see colonies.
yeahh.. that's what I thought
Thanks!!
Pour your LB plates slowly may avoid the bubbles.
If there are bubbles you can use a sterile pipette to break them before they are set.
I hope this may help.
the problem is that bubbles are at the bottom of the plates after leaving them ON at 37!
Did you put your LB plate upside down (ie. the side with the LB on top and the lid on the bottom)?
yep!
Lb contains Amp. and X-gal, could this be a reason?

I don't think so.
Just wondering whether you have mix the LB thoroughly.
If you pour the medium while it is too hot you will get bubbles after culture at 37°C.

you could also use a bunsen burner, which solves the problem a lot faster
