Splitting transfected cells: do they still overexpress my gene of interest ? - (Mar/07/2006 )

Hi,

For an experiment I would need to transfer transfected PC12 cells into a special culture system to differentiate them.
Is it fine doing that ? Will some cells still overexpress my gene of interest ?

if you've done stable transfection, there is no problem of transferring the cells in an other device. Add antibiotic to your medium if you're afraid of that...

Thanks for your reply, but it's not a stable transfection, that's why I'm asking the question.
Do you think some cells will still overexpress my gene of interest after splitting ?

generally proteins are expressed 3-4days in transient transfection.
If your cells are ok in both devices, i don't see why tehere would be no expression basically.
May you do the test with GFP transient transfected?

Good idea, I hadn't planned that but I'll definitely try it prior to the proper experiment.

you can do both if you have enough material and if the exp is not too tough and/or expensive. You'll get the result and a relative quantitation by GFP
