endothelial cells transfection - (Mar/06/2006 )

hi everyone
I'm trying to transfect porcine aortic endothelial cells. I've tried several methods including Lipofectin, Effectene, AMAXA Programs. All of them make my cells elongated, which is a big problem, because I need them for shear experiments, so I can't use them because they already changed shape.
Could anyone give me some input in this?

Our lab routinely uses gencarrier-1 for HUVEC and primary bone marrow endothelial cell transfection with great success. We did not notice any obvious cell loss or morphological change during and after transfection.
