transfection of M17 cells - (Mar/06/2006 )

Hi

Anybody experienced on transfection of BE(2)M17 cells? What is the best reagent to transiently transfect them?

We did C6, PC-12, SH-SY5Y neural and glial cell transfection using Gencarrier-2 from Epoch Biolabs. Typically, it gave aveage about 50-85% efficiency with very low cytotoxicity. BE2M17 should be similar. Good luck.

I used gencarrier-1 to transfect Hek293, caco2, colo205 cell lines, but the efficiency was not good. Caco2 was slightly better, with about 10% efficiency. The other two was less than 1%. Do you think gencarrier-2 work better than gencarrier-1?

I did not have any experience on Caco2 or Colo205 cells, but I and my colleagues (in FCCC) did many transfection on 293 cells for soluble protein expression using gencarrier-1 (which is better for common cell lines and work in the presence of serum) and consistently got more than 80% efficiency. Gencarrier-2 works better on primary and hard-to-transfect lines in our hands.

I did not have any problem with gencarrier-1 on my 293, CHO or COS-1 cells either. It always works with high efficiency and very low cytotoxicity. Be careful to store it above 4oC (not frozen)!!! and use PBS-free, serum-free and antibiotic-free solution during transfection.
