Hank's balanced salt solution, do I need it? - Will PBS do instead? (Mar/05/2006 )

Hi all,

I'm running a positive control experiment and won't need to do it more than a couple of times (fingers crossed!) and the protocol calls for HBSS with glucose. My question is, can I get away with just using PBS instead and not have to buy something that from inital searches seems rather on the expensive side. Or is there some fabulous thing about this solution which will make my labelling work a million times better. It will be on the cells for a grand total of 1 hour, and for the visualisation.

um...first of all, what are you trying to do? some sort of staining?

I would say that sometimes you can substitute one for the other, but usually the protocol calls for a given buffer for a reason. Those two buffers are pretty far apart in ingredients and in buffering capacity; I am guessing the protocol has a good reason for calling for the HBSS with glucose....I wouldn't chance it, but it's up to you.

HBSS from Fisher (I use catalog# SH3003002) is very inexpensive for 500mL... between $4-30 dollars / 500mL

I wil prepare my own HBSS... any suggestions?
