RAW 264.7 cell line transfection - (Feb/27/2006 )

Hi,

I have a problem about transfection of RAW 264.7 cells.

I'd like to transfect some CMV promoter drived constructs into the cells by using Effectene or FuGENE6 but got poor efficiency. How can I improve the transfection efficiency? It's the first time I use this cell line. Please tell me everything what a greener easily ignore.

I have worked with this cell line and I couldn't get a good transfection efficiency with Lipofectamine 2000 either. You might consider using Amaxa technology, according to their protocol you can get 65 % transfection efficiency and 74 % viability. We were also successful using Amaxa. Check this out: www.amaxa.com and search for the RAW 264.7 cell line in the database.
