unstable result with report gene analysis - (Feb/23/2006 )

i am doing promoter assay using beta-gal as transfection control. of course the results of luc activity has been normalized by comparing the beta-gal activity. i try to find out that for example. A inhibit B promoter when they are cotransfected. At the same time but another independently experiment, i am using C cotransfected with B as control. the result really puzzled me. twice i found A really inhibit B, mean the luc activity in A+B cotransfection is lower than C+B cotransfection. however i got once result showing luc activity in C+B is lower than A+B. which result i shuld believe. why the result is variant?

i am really puzzled.

do the experiment again and see which result it gives.
and make sure you did not accidentally switched the tubes/mislabel tubes during assay.
