Switching CHO cells media from high to low glucose - how would that effect cells? (Feb/14/2006 )

I am going to receive CHO cells that is cultured in DMEM high glucose. Unfortunately my lab only carry DMEM low glucose (we also have Ham F12). If I passage the cells into low glucose DMEM, what would you think would be some likely consequences? Slow growth? Stress response? Die? The cells stably transfect a protein of interest under selection antibiotic, and I do not want the cells to stop expression of that protein due to stress, etc. Thanks.

In our lab we buy DMEM with low glucose in a powder form. When we are making up 1litre of media we add 4.5g d-glucose to the media and sterile filter and BSC1 cells have been found to grow perfectly fine even when filmed by long term fluorescence. As for CHO cells, I am currently growing mine in F12/DMEM with low glucose and cells seem very happy, dividing normally with no observable cell death, so you could make a 50:50 mixture with the media you have or buy DMEM/F12 from sigma. Hope this helps
