Isolation and culture of white blood cells. - (Feb/11/2006 )

I would like to human white blood cells...stimulate them...then measure cytokine levels using the luminex. Does anyone know how many WBCs per ml I should retreive on average? I would like to do my experiment in 24 well plates. If anyone has a protocol for the isolation and stimulation of white blood cells that would be great!

Hi;
I have a protocol for isolation of monocytes ,they can be isolated from peripheral blood ,by centrifugation of diluted blood over a Ficoll-Paque density solution and harveste from the interface,after washing in PBS,culture for 2 h, after 2 h non-adherent cells should be removed by washing and the adherent monocytes should be harvested in new medium.
