over-expression - (Feb/06/2006 )

Hi

I`m trying to find a simple describe about over expression and the reasons for using it?

over expression describes itself as a higher level of expression than usual.
Applications (mains i see at time) :
-purification of protein
-exploring functions of a prot (lowering exp deserves mainly this aim, but sometimes it's better to increase the exp of target prot...)
-immortalization by expression of large T for ex or hTERT...
-mimetting overexpressions in non healthy cells

exploring functions of a protein is exactly what I was looking for. would u tell me a bit more in this aspect? what is the main point?

after that I can find some clue to search in the net.

About protein functioning determination...
Why is reducing the expression is preffered over increasing the expression...
One thing i think is its more probable that
decrease in expression---->decrease in its function
than increase in expression-----> increase in function...

Do we use both to have a confirmation....

with which organism are you working?
It is a great difference if you are planning to overexpress proteins in bacteria, plants, ...

I am going o do it in plant (Arabidopsis t.) using agrobacterium....
