transfecting primary cells - (Jan/26/2006 )

I am working with a mouse primary cell line. My experience has been that no matter which transfection reagent I try I can only acheive about 5% transfection efficiency. Can anyone tell me why primary cells are so difficult to transfect?

I don't know why transfection are thought to do with transfection reagent but I think that Qiagen sell many kind of reagents that allowed the transfection of primary cells line. Take a look on that!
