problem emerges after switch of FBS? - Anyone had this problem before? (Jan/16/2006 )

Around thanksgiving, we run out of the FBS from Cambrex. Then we use FBS from biomeda.......Our boss found the deal. The FBS from biomeda is about 100 dollars cheaper for 500ml. From then on, I could not get the results I've repeatedly seen before I switched the FBS. I've thawed new cells and the problem remains. The experiment I do are mainly transfection and luciferase assay. Anyone encounter this problem before? Any input is greatly appreciated!

Sounds like the batch of FCS is having an effect. That's why people generally test batches of FCS on their cells and in their assays before buying a big batch of FCS. Normally it's an effect on the growth of the cells. Can you not test an aliquot of another batch from your original supplier in parallel with your new batch and see if you get better results?

Agree with Ceri. There's a great batch to batch difference for FCS.

You get what you pay for. Normally, I'm all for using a cheaper provider because a lot of the big companies have obscene profit margins on a lot of the lab supply stuff (ie 5 o-rings for Gilson pipettes for nearly £30). However in the case of FBS it's well worth to pay the little bit more.

In my experience, cheaper FBS is not tested for mycoplasm. My recommendation would be to go back to your old supplier.

Yes, thank you all for suggestions. Our PI has decided to switch back to the old supplier, which we actually trust.....
