Toxicity of DMSO? - (Dec/01/2005 )

Hello,

I am presently using 10% DMSO as a cyroprotectant at -80C. The sorted cells are stored for analysis (10K Array) at a later date. Unfortunately, the amount of DNA extracted downstream is very minimal after thawing.

Two questions:

1) What happens cellularly when the DMSO is older than six months (post-expiration)?

2) What happens cellularly if more than 10% DMSO is used?

I have read of the toxicity of DMSO to particular lines of cells...but nothing is definitive.

I am trying to clear up some past issues with archived cases and any assistance would be greatly appreciated.
