Research help

i am now struggling in the field of liver, especially the hepatocytes and different kinds of immune cells in the liver.
I am interested in this work. Using the infectious model of our lab, I am sure that I can find some amazing stories in the liver.
However, I know little about liver, the structure, the experiment methods or the features of different kinds of cells.
I tried a month to stain the primary hepatocytes with fluorescence. But it turned out that the cells are strong fluorescent, and I got little information. It is really a waste of time.

Is anyone an expert in this field? So that I can learn something from you and we can discuss anything with you?

If your 1st Ab is raised in mouse, then you will use 2nd anti-mouse Ab, but if your hepatocytes is mouse, then you will be in trouble.

Because the cell will also fluorescencce with only 2nd Ab (no 1st Ab).

To overcome this problem, if you are using mouse hepatocytes
1. your 1stAb must be raised in other animal except mouse
or
2. direct labelling your 1stAb with FITC or other fluorescence.

Actually, the cells will give strong autofluorescence even you do not stain them.
