Dendritic Cells Immunotherapy - (Nov/17/2005 )

Can anybody tell me as to why Peripheral blood monocyets are chosen over bone marrow CD34 cells to induce DCs invitro for DC-Immunotherapy. Is it solely because its convienience in obtaining or are there any other reasons.
If so please cite the paper

Are you talking about mouse or human model?

I know that bone marrow is used in mouse model, while peripheral blood monocytes is used to induce DC in human clinical trial.

I know that bone marrow is used in mouse model, while peripheral blood monocytes is used to induce DC in human clinical trial.


In humans why dont they use bone marrow in clinical trials Is it because its difficult to obtain or Mo-DCs are supirior

It is bloody painful to take bone marrow from human (they scream very loud!!!!)

Thanks Minnie Mouse, I know its painful but it can be done under anaesthesia, isnt it? There are two papers saying that bone marrow DC is supirior than MO-DC but still thay use MO-DC in the clinical trial

In our case, we use PB (humans) because of insurance companies and cost. It's easier to request a blood draw (the insurance co will pay for this) than to harvest bone marrow. I'm not sure if there's another reason, though.

Not entirely sure, but I uderstand that peripheral cells are fully matured, whereas cells from BM are not. If you are looking for antigen-presenting cells, I would imagine mature, circulating cells would be a better choice than immature cells that would need to be fully differentiated.

Apart from that, the costs involved, and the insurance, ethics approvals, pain to the patient etc would be enough to want to avoid BM harvest if possible
