Help!Checking the nested Primer - P15 gene nested MSP (Nov/11/2005 )

sad.gif Hi,guys.I am a Chinese medical student.I am a newer in this area.Now.I am doing a research on the P15 gene methylation status of MOLT-4 cell line.I want to do some special,using the nested primer Msp.I get the sequence of P15 gene from my friend,the attachment.But i found that it ii different from the sequence that I found from the NCBI.Which is right?For I could not design the nested primer using the NCBI sequence,so I designed a nested Primer by putting the sequence into the Methprimer.Is that OK?Could some one help me to check the primer.Please forgive my poor English.Much thanks.

the two sequences are similar and are the mRNA sequence of CDKN2B.

I don't know why you design primers in the 5UTR region, not the promoter region.

You can first design MSP primers using the program, then pick outside primers mannually.

Hi,PCRMAN,Thank you very much for your quick reply.Yes ,they are the same sequence ,cdna of

CDKN2B,but the length is quite different.Which one should I put into the Methprimer?And the

second question is What do you mean by designing primers in the 5UTR region?Maybe this is a

stupid question,but please tell me what the 5UTR region is.The third ques tion is that I cannot pick

the outside primers mannually myself.I have been doing this ,but without any results.Please help

me !T hanks in advance.
