questions about MCF-7 and MTX - (Nov/09/2005 )

is MCF-7 sensitive to MTX? i have tried many times, but cannot get inhibition of MTX against MCF-7, even at 100 micromolar, i was wondering why? anything wrong with medium? i used DMEM

it seems that mcf-7 cell line is resistant to metothrexate...

thanks Fred_33, but this paper used mitoxantrone (MX)-selected cell line (MCF7/MX). actually paper
said the IC50 arround 22 nm. but i have tried 100 micro M, my boss suggested that i can try use the atcc recommended medium, but i do not see the point

maybe the tumor suppressor p53 is need for the action of MTX
but this protein is not detect in the cytoplasm of MCF-7 cells...

PS: serum is necessary to the induction of apoptosis of such a cell line wink.gif
