protein extraction from leaf - (Oct/31/2005 )

hi every body,

is there anybody who has used in vitro leaf for protein extraction?is the quality of protein ok?

hi,

i'm not exactly sure what you mean by an "in vitro leaf." are you referring to cultured cells derived from leaf tissue, or are you referring to extracting protein from ground leaf tissue? there are numerous protein extraction protocols for plants, and they vary quite extensively. it is important to select the buffer that best suits your system and what you intend to do with your samplesonce they are extracted. please clarify what you mean by "in vitro leaf," and i could probably give you a few recipes to try.

hi,

i want to do bioassay.actually i should infect the leaf by some pest and after that extract protein of the leaf,to check the different of the expression of CPTI gene during 2 weeks bioassay.but because its very difficalt to control the pest on the leaves on tree,can i take some leaf from tree and put the leaves in water,and doing bioassay for 2 weeks in lab?

I believe that 2 weeks in lab are a too long time for a plant explant to conserve.
I used to take leaves, flowers and petioles from field trees, sterilize them immediatly when in lab, then keep all in sterile Petri dishes for maximum 1 week at 4°C, but I didn't have to do bioassays, so maybe it's not your case.
