Importance of a freeze-thaw cycle - (Oct/23/2005 )

I would like to know the importance of a freeze-thaw cycle when it comes to homogenized tissue eg mouse heart.

What is the best way to do it, the protocol states 3 times freeze-thaw cycle?

freeze thaw cycles enhances cell shrinkage. For that, i use liquid nitrogen to freeze and a 37°C bath to thaw

you don't need to use LN2, you can use a -80C freeezer or dry ice.... freeze thaw cycles cause the formation of ice crystals in the cells which lead to lysis, slightly time concuming but useful technique
