about 3T3 and serum - (Oct/03/2005 )

Hi,

I am now culturing 3T3 cells. This cell supposed to be very strong. But my cell is in bad state sad.gif It has a lot of bubbles in side and out side of the cells. I found in some protocol, this cell should keep in 10% bovine calf serum, not fetal bovine serum. But I only have fetal serum now. Is that the cause of my poor cells?

I'm not sure what your problem is but 3T3's will grow Better in fetal compared to calf serum.

you can try to decomplement your serum by heating it at 56° for 30'

Recently we were facing the same problem. So we have been testing 6 different batches of 3T3 cells (they all have a different morphology) and it turned our that they all grow fine on Newborn Calf Serum 9% (heat inactivated) but very bad on Fetal Calf serum (9%)

I have seen this also and it has varied depending upon where I got my cells from. It is amazing that 3T3 cells seem to vary so much depending upon the lab you got them from, but this seems to be the case.

All the different 3T3s not only differ in their serum requirements. Also in their state of transformation, stress response, growthrate etc.

I'll ditto that statement. That is why you need to take 3T3 transformation studies with a grain of salt or at least know where they got "their" 3T3 cells from
