plasmid construct problem - (Aug/10/2009 )

hi,everyone.I am a new in plasmid construct. Is there anyone can recommend me some books to read or some tips can give to me! I want to know from the beginning:1.primer design for ORF clone. 2. tips about plasmid enzyme digestion and ligation. Just like a course from beginning.

what would you like to know? typically, for basic info, you can just browse the net, or any Molecular Biology textbook will give you a general feel.

Tips:

Make sure your primers will keep the sequence in frame.

For digests, choose single site cutters so that you don't cut the plasmid more than once. Also ensure that your insert has complementary ends.
